Across TCGA pan-cancer cohorts, TLR4 RNA differs between tumor and matched normal tissue in 15 of 18 cancer types tested, making tumor–normal expression one of TLR4’s most consistent transcriptional readouts.

The strongest signal is observed in lung adenocarcinoma (LUAD), where TLR4 RNA is repressed in tumor relative to normal tissue. In most cancer types TLR4 is over-expressed in tumor, although a few such as LUAD and LUSC show the opposite, repressed pattern.

LUAD, LUSC, and BRCA are the cancer types where TLR4 tumor–normal differential expression is most reproducible.
