Across TCGA pan-cancer cohorts, SMPD3 RNA is linked to patient survival in 26 of 34 cancer types, making it the most broadly survival-associated SMPD3 data layer compared with 9 for mutation status and 3 for mass-spec protein.

The strongest signal is observed in head and neck squamous cell carcinoma (HNSC), where higher SMPD3 RNA is associated with better disease-free survival. In most high-consensus cancer types, elevated SMPD3 expression acts as an unfavorable survival marker, although some lineages such as HNSC and LGG show a favorable association.

HNSC, LGG, and UCEC are the cancer types where SMPD3 RNA most reproducibly stratifies survival.
