Across TCGA pan-cancer cohorts, SMAD7 RNA is linked to patient survival in 22 of 34 cancer types, making it the most broadly survival-associated SMAD7 data layer compared with 8 for mutation status and 1 for mass-spec protein.

The strongest signal is observed in lung squamous cell carcinoma (LUSC), where higher SMAD7 RNA is associated with worse overall survival. In most high-consensus cancer types, elevated SMAD7 expression acts as an unfavorable survival marker, although some lineages such as LGG and KIRC show a favorable association.

LUSC, LGG, and ACC are the cancer types where SMAD7 RNA most reproducibly stratifies survival.
