Across TCGA pan-cancer cohorts, RUNX2 RNA differs between tumor and matched normal tissue in 15 of 18 cancer types tested, making tumor–normal expression one of RUNX2’s most consistent transcriptional readouts.

The strongest signal is observed in thyroid carcinoma (THCA), where RUNX2 RNA is more highly expressed in tumor relative to normal tissue. In most cancer types RUNX2 is over-expressed in tumor, although a few such as PRAD show the opposite, repressed pattern.

THCA, LUAD, and KIRC are the cancer types where RUNX2 tumor–normal differential expression is most reproducible.
