Across TCGA pan-cancer cohorts, PENK RNA is linked to patient survival in 25 of 34 cancer types, making it the most broadly survival-associated PENK data layer compared with 5 for mutation status and 2 for mass-spec protein.
The strongest signal is observed in uterine carcinosarcoma (UCS), where higher PENK RNA is associated with better disease-free survival. In most high-consensus cancer types, elevated PENK expression acts as an unfavorable survival marker, although some lineages such as UCS and HNSC show a favorable association.
UCS, HNSC, and UVM are the cancer types where PENK RNA most reproducibly stratifies survival.
