Across TCGA pan-cancer cohorts, MC4R Mutation is linked to patient survival in 7 of 34 cancer types, making it a survival-associated MC4R data layer compared with 20 for mass-spec protein.

The strongest signal is observed in breast invasive carcinoma (BRCA), where higher MC4R Mutation is associated with worse overall survival. In most high-consensus cancer types, elevated MC4R expression acts as an unfavorable survival marker, although some lineages such as UCEC and HNSC show a favorable association.

BRCA, DLBC, and SKCM are the cancer types where MC4R Mutation most reproducibly stratifies survival.
