Across TCGA pan-cancer cohorts, MC4R RNA differs between tumor and matched normal tissue in 7 of 18 cancer types tested, making tumor–normal expression one of MC4R’s most consistent transcriptional readouts.

The strongest signal is observed in stomach adenocarcinoma (STAD), where MC4R RNA is repressed in tumor relative to normal tissue. In most cancer types MC4R is over-expressed in tumor, although a few such as STAD and KICH show the opposite, repressed pattern.

STAD, LUAD, and KICH are the cancer types where MC4R tumor–normal differential expression is most reproducible.
