Across TCGA pan-cancer cohorts, SMPDL3B RNA is linked to patient survival in 26 of 34 cancer types, making it the most broadly survival-associated SMPDL3B data layer compared with 5 for mutation status and 4 for mass-spec protein.
The strongest signal is observed in head and neck squamous cell carcinoma (HNSC), where higher SMPDL3B RNA is associated with better overall survival. In most high-consensus cancer types, elevated SMPDL3B expression acts as an unfavorable survival marker, although some lineages such as HNSC and SKCM show a favorable association.
HNSC, UVM, and SKCM are the cancer types where SMPDL3B RNA most reproducibly stratifies survival.
