Across TCGA pan-cancer cohorts, SMAD4 RNA is linked to patient survival in 26 of 34 cancer types, making it the most broadly survival-associated SMAD4 data layer compared with 6 for mutation status and 3 for mass-spec protein.

The strongest signal is observed in adrenocortical carcinoma (ACC), where higher SMAD4 RNA is associated with worse disease-free survival. In most high-consensus cancer types, elevated SMAD4 expression acts as an unfavorable survival marker, although some lineages such as KIRC and PAAD show a favorable association.

ACC, KIRC, and PAAD are the cancer types where SMAD4 RNA most reproducibly stratifies survival.
