Across TCGA pan-cancer cohorts, SMAD3 Mutation is linked to patient survival in 8 of 34 cancer types, making it a survival-associated SMAD3 data layer compared with 21 for mass-spec protein and 5 for mass-spec protein.
The strongest signal is observed in kidney chromophobe (KICH), where higher SMAD3 Mutation is associated with worse disease-free survival. In most high-consensus cancer types, elevated SMAD3 expression acts as an unfavorable survival marker, although some lineages such as BLCA show a favorable association.
KICH, LUAD, and LGG are the cancer types where SMAD3 Mutation most reproducibly stratifies survival.
