Across TCGA pan-cancer cohorts, SIX3 RNA is linked to patient survival in 26 of 34 cancer types, making it the most broadly survival-associated SIX3 data layer compared with 3 for mutation status.

The strongest signal is observed in acute myeloid leukemia (LAML), where higher SIX3 RNA is associated with better disease-free survival. In most high-consensus cancer types, elevated SIX3 expression acts as an unfavorable survival marker, although some lineages such as LAML and BLCA show a favorable association.

LAML, LIHC, and LGG are the cancer types where SIX3 RNA most reproducibly stratifies survival.
