Across TCGA pan-cancer cohorts, SH2B1 RNA differs between tumor and matched normal tissue in 9 of 18 cancer types tested, making tumor–normal expression one of SH2B1’s most consistent transcriptional readouts.

The strongest signal is observed in liver hepatocellular carcinoma (LIHC), where SH2B1 RNA is more highly expressed in tumor relative to normal tissue. In most cancer types SH2B1 is over-expressed in tumor, although a few such as THCA and UCEC show the opposite, repressed pattern.

LIHC, THCA, and KIRP are the cancer types where SH2B1 tumor–normal differential expression is most reproducible.
