Across TCGA pan-cancer cohorts, POLR1A RNA differs between tumor and matched normal tissue in 14 of 18 cancer types tested, making tumor–normal expression one of POLR1A’s most consistent transcriptional readouts.

The strongest signal is observed in colon adenocarcinoma (COAD), where POLR1A RNA is more highly expressed in tumor relative to normal tissue. In most cancer types POLR1A is over-expressed in tumor, although a few such as THCA show the opposite, repressed pattern.

COAD, HNSC, and STAD are the cancer types where POLR1A tumor–normal differential expression is most reproducible.
