Across TCGA pan-cancer cohorts, PNPLA2 RNA is linked to patient survival in 27 of 34 cancer types, making it the most broadly survival-associated PNPLA2 data layer compared with 2 for mutation status and 6 for mass-spec protein.

The strongest signal is observed in breast invasive carcinoma (BRCA), where higher PNPLA2 RNA is associated with better overall survival. In most high-consensus cancer types, elevated PNPLA2 expression acts as an unfavorable survival marker, although some lineages such as BRCA and SCLC show a favorable association.

BRCA, LGG, and ACC are the cancer types where PNPLA2 RNA most reproducibly stratifies survival.
