Across TCGA pan-cancer cohorts, PKN1 RNA differs between tumor and matched normal tissue in 15 of 18 cancer types tested, making tumor–normal expression one of PKN1’s most consistent transcriptional readouts.

The strongest signal is observed in kidney renal clear cell carcinoma (KIRC), where PKN1 RNA is more highly expressed in tumor relative to normal tissue. In most cancer types PKN1 is over-expressed in tumor, although a few such as LUSC and LUAD show the opposite, repressed pattern.

KIRC, KIRP, and COAD are the cancer types where PKN1 tumor–normal differential expression is most reproducible.
