Across TCGA pan-cancer cohorts, IGHV3-16 RNA is linked to patient survival in 21 of 34 cancer types, making it the most broadly survival-associated IGHV3-16 data layer compared with 1 for mutation status and 2 for mass-spec protein.

The strongest signal is observed in head and neck squamous cell carcinoma (HNSC), where higher IGHV3-16 RNA is associated with better overall survival. In most high-consensus cancer types, elevated IGHV3-16 expression acts as an unfavorable survival marker, although some lineages such as HNSC and SKCM show a favorable association.

HNSC, SKCM, and LUAD are the cancer types where IGHV3-16 RNA most reproducibly stratifies survival.
